You can bicycle on any of the paved roads in the park, including 33-mile Rim Drive and on Grayback Drive, an unpaved, un-motorized road off of Pinnacle Drive. Be careful on the narrow, winding roads, as there are lots of blind corners and steep hills that may prevent cars from spotting you ahead of time. The best bet is to start early in the morning, when car traffic and wind speeds are low.
